Fast, Safe Neutralization of Anti‑CD38 Interference

DaraEx & DaraEx plus

Antibody based drugs can disturb serological blood group testing since the relevant antigens cannot be examined due to the therapeutic antibodies. This may lead to (false‑positive) reactions in the indirect antiglobulin test (IAT) — making routine serological testing challenging and time‑consuming.

Using the standard Dithiothreitol (DTT) technique can reduce interference, but with major drawbacks: destruction of Kel antigens, hemolysis, and complex workflows.

DaraEx (For Research Use Only) and the CE‑IVD marked DaraEx plus offer a fast, safe, and e+asy solution to inhibit the agglutination effect of anti-CD38 in IAT without side effects.

Why DaraEx / DaraEx plus?

  • Simple integration into routine serology
  • Express protocol — no pre-incubation of cells required!
  • Specific inhibition of anti‑CD38 antibodies
  • Preserves blood group antigens — no side effects like destruction of blood group antigens or hemolysis
  • No need for anti‑idiotypic antibodies or soluble CD38 protein
  • Process control available (Dara-PC)
  • 15‑month shelf life
  • Now also available as CE-IVD product DaraEx plus (NEW)

How It Works

DaraExbild1_540x540px.png

DaraEx / DaraEx plus is a Fab‑fragment of an anti-CD38 antibody that masks CD38 on the erythrocytes surface, preventing therapeutic anti‑CD38 antibodies from binding.

This enables accurate antibody screening, identification, and cross‑matching — all with one express protocol.

No preincubation of DaraEx / DaraEx plus with the test cells necessary:

» to 1 volume of cells (0.8 %) 0.2 volume of DaraEx / DaraEx plus is added
» e.g. for 50 μL cells 10 μL DaraEx / DaraEx plus is required

Compatible with standard gel card systems (Bio‑Rad ID‑System, ORTHO MTS).

Left: The anti-CD38 antibody (green) in a patient sample binds to CD38 (yellow) on red blood cells, causing agglutination by anti-human globulin (purple) in the IAT.
Right: In the Antigen Masking Indirect Antiglobulin Test (AMIAT), a specific antigen on the red blood cell surface is masked before anti-human globulin is added. With DaraEx / DaraEx plus (blue)—a Fab‑fragment of an anti‑CD38 antibody—CD38 is masked, preventing therapeutic anti‑CD38 antibodies from binding.
